When incorporated into the protein of interest, epitope tags make protein detection and identification very robust and easy. The number of epitope tags has increased considerably, and tags like DYKDDDDK, His, and HA.11 are commonly used in cell biology research labs. BioLegend now offers directly-conjugated epitope tag antibodies for easy surveillance of the tagged fusion protein in immunofluorescence and intracellular flow cytometry assays.
Alexa Fluor® 488 Alexa Fluor® 594 Alexa Fluor® 647
HeLa cells transfected with multi-tag plasmid were fixed with cold methanol for 15 minutes at -20°C and then blocked with 5% FBS for 60 minutes. Cells were stained with either Alexa Fluor® 488 (left, green), Alexa Fluor® 594 (middle, red), or Alexa Fluor® 647 anti-His antibody (right, red), and incubated overnight at 4°C. Nuclei were counterstained with DAPI (blue).
Brilliant Violet™ 421 Alexa Fluor® 488 Alexa Fluor® 594
CHO cells stably transfected with HA tag were fixed, permeabilized, and blocked. Then the cells were intracellularly stained with Brilliant Violet™ 421 (left, blue), Alexa Fluor® 488 (middle, green), or Alexa Fluor® 594 anti-HA.11 Epitope Tag Antibody (right, red) overnight at 4°C. For the left image, nuclei were counterstained with DRAQ5™ (red). For the middle and right images, cells were counterstained with DAPI (blue).
Alexa Fluor® 594 Alexa Fluor® 647 APC
HeLa cells transiently transfected with DYKDDDDK tag fused protein were fixed, permeabilized, and blocked. Then the cells were intracellularly stained with either Alexa Fluor® 594 (left, red) or Alexa Fluor® 647 anti-DYKDDDDK Tag antibody (middle, red). Nuclei were counterstained with DAPI (blue). HEK-293T cells transfected with plasmid encoding a DYKDDDDK-tagged protein were fixed, permeabilized, and then stained with anti-DYKDDDDK tag (clone L5) APC (filled histogram) or rat IgG2a, κ APC isotype control (open histogram).
