Total cell lysates (15 µg total protein) from NTERA-2, HeLa, HEL, and Jurkat cells were resolved by 4-20% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 0.1 µg/mL (1:5000 dilution) of Purified anti-IRF2 Antibody, clone 13B2A38, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse-IgG Antibody (Cat. No. 405306). Direct-Blot™ HRP anti-β-Actin Antibody (Cat. No. 664804) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular Weight marker. Predicted expression data was obtained from Human Protein Atlas. Cell lysates were loaded in order of increasing IRF2 expression levels (source: Human Protein Atlas).

Description

IRF2 is a member of the IRF family of transcription factors, mainly functioning as a transcriptional repressor. It binds to the interferon-sensitive response elements (ISREs) and competes for binding sites with the other IRF transcription factors, such as IRF1 and IRF9. Mice deficient in IRF2 spontaneously develop skin inflammation due to excessive activation of CD8+ T lymphocytes. Further, the severely reduced number of mature NK cells in IRF2-deficient mice suggests that IRF2 is required for normal NK cell development. IRF2 contains a latent activation domain that can act as a transcriptional activator and has been revealed to positively regulate Histone H4 and VCAM-1 gene expression.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

During PD testing for western blot application, this clone cross-reacted with a ~50 kDa protein of unknown origin when used at concentrations exceeding 0.1 µg/mL.
