    Total cell lysates (15 µg total protein) from HEL, A431, A549 and NIH/3T3 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 0.5 µg/mL (1:1000 dilution) of Purified anti-LASP1 Antibody, clone 8C6, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker. Cell lysates were loaded in order of increasing LASP1 mRNA expression levels (source: Human Protein Atlas).

    Immunofluorescence of HepG2 cells with (A) mouse IgG1, κ isotype control (Negative, Cat. No. 401402) or (B-D) LASP1 antibody (Clone 8C6). Alexa Fluor® 488 (Green) Goat anti-Mouse IgG (Cat. No. 405319) was used as secondary antibody. Nuclei were counterstained with DAPI (Blue, Cat. No. 422801). The image was captured with a 60X objective using KEYENCE BZ-X700 fluorescence microscope. Exposure time (Seconds) for (A-D) is 1/15. Concentrations for (A, B) is 10 µg/ml, (C) is 4 µg/ml and (D) is 2 µg/ml.

Description

Lasp-1 (LIM and SH3 domain protein 1) is an actin-binding phosphoprotein expressed in a wide range of cell types rich in F-actin. Lasp-1 is believed to play a role in the reorganization of the actin cytoskeleton, and localizes to sites of cytoskeletal assembly in response to signalling inputs. Lasp-1 binding partners, in addition to actin, include the endocytic proteins dynamin 2, Hip1r and clathrin, and proteins involved in focal adhesion including zyxin and krp1. The functions of Lasp-1 protein are not well defined, but evidence suggests roles in cell migration, tumor formation, and membrane remodeling in the regulation of ion transport.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a dilution range of 1:100 - 1:200 is suggested. For Western blotting, the suggested use of this reagent is 0.5 - 1.0 µg per mL (1:1000 - 1:2000 dilution). For immunocytochemistry, a concentration of 10.0 μg/mL (1:100 dilution) is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody is effective in immunoblotting (WB) and immunohistochemistry (ICC).
