B-Cell Receptor Pathway
The B cell receptor complex is comprised of membrane-bound immunoglobulin (Ig) and Igα/β proteins that have immunoreceptor tyrosine activation motifs (ITAM). The signal is initiated when extracellular antigen binds the membrane Ig, which promotes Lyn phosphorylation of the Igα/β proteins. Syk is then recruited and activated by phosphorylated Igα/β, and goes on to phosphorylate BLNK. BLNK acts as a scaffold from which multiple signaling pathways become activated. The outcome of B cell receptor signaling can often depend on signals from other receptors engaged by cytokines or ligands on other cell types, such as helper T cells and/or antigen presenting cells (APCs). This can result in the initiation of many different cellular processes, including B cell differentiation and maturation.
