Western blot of purified anti-ApoER2 (LRP8) antibody (clone 25G5). Lanes 1, 5: Molecular weight marker; Lanes 2, 6: 40 µg of mouse brain lysate; Lanes 3, 7: 40 µg of rat brain lysate; Lanes 4, 8: 20 µg of SH-SY5Y cell lysate. The blots were incubated with 2 µg/mL of the primary antibody or Rat IgG2a isotype control overnight at 4°C, followed by incubation with HRP-labeled goat anti-rat IgG (Cat. No. 405405). Enhanced chemiluminescence was used as the detection system.

Description

LDL Receptor Related Protein 8 (LRP8), also known as ApoER2, is a receptor for members of low-density lipoproteins. LRP8 interacts with its ligand reelin, and triggers reelin signaling to regulate cellular processes such as neuronal cell migration and long-term potentiation of mature neurons. LRP8 also interacts with and downregulates APP endocytosis. This interactions leads to increased amyloid beta production, demonstrating a major role for LRP8 in the pathogenesis of  Alzheimer’s disease.
