Immunofluorescence staining of anti-Neurofilament H (NF-H), Nonphosphorylated antibody (clone SMI 32) on formalin-fixed paraffin-embedded (FFPE) tissue section from rat cerebellum. After antigen retrieval using Retrieval-ALL Antigen Unmasking System 3, the tissue was blocked with 5%NGS + 1%BSA (2nd image) or Normal Serum Block (1st image) for 30 min at room temperature, and then incubated with SMI 32 at 5 µg/mL (diluted in 5%NGS+1%BSA or Normal Serum Block, respectively) overnight at 4°C. Images were captured with a 40X objective (Scale Bar: 20 µm).

The optimal working dilution should be determined for each specific assay condition.

Application Notes

Normal goat serum is used as a blocking solution in immunohistochemistry. It is effective in reducing non-specific binding of proteins. For example, Normal Serum Block prevents the secondary antibody from cross reacting with endogenous immunoglobulins in the tissue. The use of normal serum block before the application of primary antibody also eliminates Fc receptor binding of both the primary and secondary antibody.

The Normal Serum Block (cat#s 927501, 927502 ,  927503) is supplied as a blue-colored solution for easy identification when working with multiple IHC reagents. Internal testing has indicated that this product does not interfere with immunofluorescent signals, nor did it contribute to autofluorescence in microscopy work.
