Total tissue lysates (15 µg total protein) from rat brain (Lane 1), mouse brain (Lane 2), human brain (Lane 3), and human kidney (Lane 4, negative control) were resolved by 4-12 % Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 μg/mL of purified anti-GABABR2 recombinant (clone QA19A84) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at 1:3000 dilution. Direct-Blot™ HRP anti-β actin (Cat. Description

Gamma-aminobutyric acid, B [GABAB] receptors are located in the midbrain and help facilitate inhibitory signaling in the central nervous system. GABAB receptors exist as heterodimers, comprising of two subunits: GABABR1 and GABABR2 with 7 transmembrane domains, neither of which is fully functional when expressed alone. The GABABR1 inhibits neurotransmitter activity via binding γ-aminobutyric acid (GABA), following G protein-coupled pathway activation through interacting with GABABR2. Subsequently, this signaling cascade changes ion channel activity and induces adenylyl cyclase to interfere synaptic responses. A decrease in GABABR2 functional efficacy is implicated in several diseases, such as Rett syndrome and epileptic encephalopathy.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

It is recommended to boil cell or tissue lysates at 70°C for western blot.

Reactivity of clone QA19A84 with human GABABR2 is observed but is weaker than reactivity with mouse or rat GABABR2.
