IHC staining of purified anti-α smooth muscle actin (clone 1A4) on formalin-fixed paraffin-embedded human kidney tissue. Following antigen retrieval using 1X Tris-Buffered Saline (final concentration 0.05M) with Tween-20 (Cat. No. 925501), the tissue was incubated without (panels A and C) and with (panels B and D) 5 µg/mL of primary antibody followed by incubation with Alexa Fluor® 647 goat anti-mouse IgG antibody (Cat.     HeLa cells were fixed with ice-cold methanol and blocked with 5% FBS for 1 hour at room temperature.     Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.25 µg/mL of purified anti-α smooth muscle actin (clone 1A4) overnight at 4°C. Description

Cytoplasmic actins, which belong to the microfilament system of cytoskeleton proteins, are some of the most conserved eukaryotic proteins expressed in mammals. α-smooth muscle actin is widely expressed in contractile cells, most notably the mural cells that line arteries and arterioles. Overexpression of α-smooth muscle actin is a hallmark of fibrosis, a thickening or scarring of damaged tissue.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration of 1.25 μg/mL is recommended. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

When used in ICC testing, we recommend fixing and permeabilizing with ice-cold methanol.
