    Jurkat cells (negative cell line, panels A and B) and HeLa cells (positive cell line, panels C and D) were fixed and permeabilized with 100% ice-old methanol for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with 5 μg/mL of purified anti-EphB4 (clone W20295K), followed by incubation with Alexa Fluor® 594 goat anti-rat IgG (Cat. No. 405422) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801, panels A and C). Images were captured with a 60X objective. Scale bar: 50 µm

Description

Ephrin type-B receptor 4 (EphB4) is a member of the Eph family of receptor tyrosine kinases that bind to the Ephrin family of ligands. Specifically, EphB4 binds to Ephrin B2, in a process involving direct cell-to-cell interaction, to initiate signaling pathways that influence cell behavior such as differentiation, adhesion and migration.  EphB4 has been shown to be involved in heart morphogenesis and angiogenesis, and in the tumorigenesis of cancers such as gliomas, breast and prostate cancer.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 1.0 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This product is not recommended for use in IHC-P.

This product does not recognize mouse EphB4 in WB.

For ICC, we recommend fixation/permeabilization with 100% ice-cold methanol. We do not recommend using 4% PFA as a fixative.
