    HeLa cells were fixed with 4% paraformaldehyde for 30 minutes, permeabilized with 0.5% Triton X-100 for 15 minutes, and blocked with 5% FBS for 30 minutes. Then the cells were intracellularly stained with 2.5 µg/mL purified anti-human BMI1 (clone W20031H) overnight at 4°C followed by Alexa Fluor® 555 goat anti-rat IgG (red) for one hour at room temperature. Cells were counterstained with Flash Phalloidin Green 488 (green).

    15 µg total cell lysate from U937 cells (lane1), HEL cells (lane 2), NTERA-2 (lane 3), HeLa cells (lane 4,), were resolved by electrophoresis (4-12% Bis-Tris gel), transferred to a PVDF membrane, and probed with 1 µg/mL purified anti-human BMI1 (clone W20031H). Proteins were visualized using an HRP goat anti-rat IgG (Cat. No. 405405) and chemiluminescence detection. Direct-Blot™ HRP anti-GAPDH was used as a loading control.

Description

BMI1 is a ring finger protein that is a major component of the polycomb group complex 1 (PRC1). Acting via chromatin remodeling and modification of histones, PRC1 maintains the transcriptionally repressive state of multiple regulatory genes involved in embryonic development and self-renewal in somatic stem cells. BMI1 is essential for the self-renewal of hematopoietic stem cells and neuronal stem cells, in part via repression of genes involved in senescence. Aberrant expression of BMI1 is associated with many cancers and resistance to chemotherapy.

Recommended Usage

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. For immunocytochemistry, a concentration of 2.5 μg/mL is recommended. For western blotting, the suggested use of this reagent is 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
