Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of purified anti-RSK1 (clone 6B9-D6-F8) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Western-Ready™ ECL Substrate Plus Kit (Cat. No. 426317) was used as a detection agent. Lane M: Molecular weight marker

Description

Ribosomal protein S6 kinase α1 (RPS6KA1), most commonly known as RSK1, is a serine/threonine kinase that acts downstream of ERK/MAPK signaling. Upon activation, RSK1 translocates to the nucleus where it controls the activity of several important transcription factors, such as CREB1 and c-FOS. Additionally, RSK1 directly controls translation through phosphorylation of the transcriptional protein RPS6 and EIF4B. Through these roles, RSK1 is involved in the control of cellular growth and proliferation, and genetic variants of RSK1 have been implicated in multiple cancer types.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
