Whole cell extracts (15 µg protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to PVDF and probed with 1.0 µg/mL (1:500 dilution) purified anti-TEC antibody (clone W19291E) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin antibody (Cat. No. 643807) was used as a loading control at a 1:10000 dilution (lower). Lane M: Molecular weight marker.

The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with clone W19291E followed by a light chain-specific HRP conjugated secondary anti-rat antibody. Description

TEC is a part of the non-receptor protein tyrosine kinases subfamily which includes Bmx, Itk/Tsk/Emt, Rlk/Txk, and Btk proteins. As a non-receptor protein kinase, TEC contributes to signaling from many receptors and acts as a signal transducer in multiple downstream pathways, this includes the regulation of the actin cytoskeleton. The enzyme’s primary location in mammalian tissues is the plasma membrane. It is involved in regulating the development, function and differentiation of conventional T-cells and nonconventional NKT-cells.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.5 - 1.0 µg/mL. For immunoprecipitation, the suggested use of this reagent is 2.5 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone was tested for ICC using HeLa cells treated with control siRNA or siRNA targeting TEC. While staining was observed, it was not responsive to TEC protein depletion using siRNA, indicating the staining was non-specific. We therefore do not recommend this clone for ICC.
