Description

TOX is a member of the high-mobility group (HMG) box superfamily of DNA-binding proteins and is originally identified as the thymic transcript expressed in CD4+CD8+ double positive T-cells. TOX is also defined as part of a small subfamily of transcription factors consisting of TOX2, TOX3, and TOX4, which share conserved HMG-box sequences. TOX is highly upregulated in thymocytes undergoing positive selection and is required for the differentiation and development of various T-cell subsets, such as CD4+ T cells, regulatory T cells, and natural killer T cells (NKTs). TOX-deficient mice fail to develop lymph nodes and have significantly diminished numbers of CD4+ T-cells, NK cells, and ILC cells.

Recommended Usage

Each lot of this antibody is quality control tested by immunocytochemistry. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 594 has an excitation maximum of 590 nm, and a maximum emission of 617 nm.

Application Notes

The immunogen used to generate clone 6E6D03 is the N-terminus of Tox1, which is highly conserved among Tox family member proteins. Hence, this clone may recognize multiple Tox family members. Predicted MW for Tox family members: Tox 1 is 57.5 kD, Tox 2 is 49, 52, 53, 55 kD, Tox 3 is 63 kD and Tox 4 is 64, 66 kD.
