Description

Six interleukin-17 (IL-17)-family (IL-17A-IL-17F) and five IL-17 receptor (IL-17R)-family (IL-17RA-IL-17RE) members have been reported in mammalian cells. IL-17RA (known as IL-17R, IL-17AR, CD217) is a 128-158 kD type I transmembrane glycoprotein. IL-17R binds to IL-17A (also known as IL-17, and CTLA-8, is a hallmarker of Th17 cells) and IL-17F and mediates the proinflammatory signals through interaction with IL-17RC or IL-17RD. IL-17R has broad tissue distribution, including fibroblasts, epithelial cells, endothelial cells, stromal cells, B cells, T cells, NK cells, monocytes and granulocytes.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

* PE/Dazzle™ 594 has a maximum excitation of 566 nm and a maximum emission of 610 nm.

Structure
IL17A homodimer preferentially drives the formation of IL17RA-IL17RC heterodimeric receptor complex. IL17A homodimer adopts an asymmetrical ternary structure with one IL17RA molecule, allowing for high affinity interactions of one IL17A monomer with one IL17RA molecule (via D1 and D2 domains), while disfavoring binding of a second IL17RA molecule on the other IL17A monomer (By similarity)

Highly expressed in T cells and macrophages and in B-1a B cells and at a lower extent in B-1b and B-2 B cells (at protein level)

Function
Receptor for IL17A and IL17F, major effector cytokines of innate and adaptive immune system involved in antimicrobial host defense and maintenance of tissue integrity; cytokine binding triggers homotypic interaction of IL17RA and IL17RC chains with TRAF3IP2 adapter, leading to TRAF6-mediated activation of NF-kappa-B and MAPkinase pathways, ultimately resulting in transcriptional activation of cytokines, chemokines, antimicrobial peptides and matrix metalloproteinases, with potential strong immune inflammation
