Whole cell extracts (15 µg total protein per lane) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with the 1 µg/mL of Purified anti-Dynamin-2 (clone W22024F) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:5000 dilution. Direct-Blot™ HRP anti-β-actin (Cat.     HeLa cells (mock) (panels A and B) and Dynamin-2 knockdown cells (KD) (panels C and D) were fixed with Fixation Buffer (Cat. No. 420801) for 10 minutes, permeabilized with ice-cold methanol, and then blocked with 5% FBS for 1 hour at room temperature, respectively. Cells were then stained with Purified anti-Dynamin-2 (clone W22024F), followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 Goat anti-rat IgG (Cat. No. 405422) for 1 hour at room temperature.     293T cells which were untransfected (filled histogram) or transfected with a knockdown shRNA against Dynamin-2 (open histogram) were fixed with Fixation Buffer (Cat. No. 420801) then permeabilized with True-Phos™ Perm Buffer (Cat. No. 425401). Cells were then stained with either Purified anti-Dynamin-2 (solid line histograms) or purified rat IgG2a, κ isotype control (Cat. Description

Dynamin-2 (DNM2) belongs to the dynamin family. It consists of GTPase domain (G-domain) at the N-terminus, middle domain, the lipophilic pleckstrin homology [PH] domain, GTPase effector domain (GED), and proline rich domain (PRD) at the C-terminus. The PH domain interacts with membrane phosphoinositides, while PRD domain interacts with SH3 domain-containing proteins. Dynamin-2 is a large mechanochemical GTPase involved in various cellular processes such as endocytosis, apoptosis, cytokinesis, phagocytosis, neuronal morphology, axonal growth, cell migration, cell membrane trafficking and cytoskeletal regulation. The studies indicate that mutations in Dynamin-2 cause centronuclear myopathy (CNM), Charcot-Marie-Tooth disease dominant intermediate type B (CMTDIB), hereditary spastic paraplegia and lethal congenital contractures syndrome type 5 (LCCS5). Additionally, numerous studies have implicated Dynamin-2 overexpression in several types of cancer and metastasis.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/mL is recommended. For immunohistochemistry, a concentration range of 0.2 - 5.0 µg/mL is suggested. For flow cytometric staining, the suggested use of this reagent is ≤ 0.0625 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

The clone does not cross-react with mouse in western blotting (WB).

For immunocytochemistry (ICC), we recommend to use 4% PFA fixation followed by permeabilization with ice cold-methanol, or ice-cold methanol fixation.

For immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), antigen retrieval with Citrate Buffer (Cat. No. 420901) or Tris-EDTA pH 9.0 is recommended.

For intracellular staining for flow cytometry (ICFC), the recommended dosage is 0.0625 μg per test using intracellular staining buffer (Cat. No. 421002), Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803), True-Phos™ Perm Buffer (Cat. No. 425401) and True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401).

Microtubule-associated force-producing protein involved in producing microtubule bundles and able to bind and hydrolyze GTP. Plays a role in the regulation of neuron morphology, axon growth and formation of neuronal growth cones. Plays an important role in vesicular trafficking processes, in particular endocytosis. Dynamin-2 is also involved in cytokinesis.
