Human CD3+ T cells were isolated from healthy human PBMCs using the MojoSort™ Human CD3 T Cell Isolation Kit (Cat. No. 480021). The CD3+ T cells were expanded with Human CD3/CD28 T Cell Activation Beads. At day 3 post-activation, the beads were removed using a MojoSort™ Magnet (Cat. No. 480019) while the cells remained in culture in the presence of IL-2 (Cat. No. 589102).

Cell debris, dead cells, and doublets were excluded from analysis.

Description

The Human CD3/CD28 T Cell Activation Beads allow for convenient in vitro activation of human T cells without the need for antigen-presenting cells. Human T cells are stimulated with anti-CD3 and anti-CD28 antibodies coated on superparamagnetic microbeads. Simply add the beads directly to isolated T cells or to PBMCs at a 1:1 particle-to-cell ratio for robust T cell activation. The antibody-coated beads can be removed from the cells by the use of a magnetic separator. Some of the downstream applications include functional assays, gene expression analysis, and phenotypic characterization.

Preparation
The antibodies were purified by affinity chromatography, and conjugated with magnetic microbeads under optimal conditions.
Storage & Handling
All components should be stored undiluted between 2°C and 8°C.
Recommended Usage

Use Human CD3/CD28 T Cell Activation Beads at a 1:1 particle-to-cell ratio.

Application Notes
