Whole cell extracts (15 µg total protein) from indicated cell lines were resolved in 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-ZO-3 (clone W22143C) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker.

    LN-319 cells (negative cell line, panel A) and MCF-7 cells (positive cell line, panel B) were fixed with Fixation Buffer (Cat. No. 420801) and permeabilized with 0.5% Triton X-100, and blocked with 5% FBS. The cells were stained with Purified anti-ZO-3 (clone W22143C) overnight at 4°C, followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective. Scale bar: 50 µm

Description

ZO-3 is a member of the membrane-associated guanylate kinase-like (MAGUK) protein family. Together with ZO-1 and ZO-2, they act as scaffold proteins that link transmembrane proteins in the tight junctions such as claudins, junctional adhesins and occludins to the actin cytoskeleton forming a boundary between the apical and basolateral plasma membrane on epithelial cells. The importance of this complex of proteins is to maintain tight junctions and limit the paracellular movement of substances. In addition, ZO-3 sequesters Cyclin D1 avoiding its degradation during mitosis and enabling S-phase transition. Celiac Disease 1 has been associated with ZO-3.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Function
