Total cell lysates (15 µg protein) from NIH3T3 (lane 1), HeLa (lane 2) and HaCaT (lane 3) cells were resolved by electrophoresis (4-12% Bis-Tris gel), transferred to nitrocellulose, and probed with 0.5 µg/mL (1:1000 dilution) of purified anti-BCL10 antibody (clone P84F8) (upper blot). Proteins were visualized by chemiluminescence detection using a 1:3000 diluted goat anti-mouse-IgG secondary antibody conjugated to HRP for anti-BCL10 antibody or 1:5000 diluted Direct-Blot HRP anti-β-Actin antibody, clone 2F1-1(lower blot). Lane M: Molecular weight ladder.

Description

BCL10 (B cell lymphoma 10), together with CARMA1 (CARD11) and lymphoid tissue lymphoma-translocation gene 1 (MALT1) are signaling proteins. They have a key role in antigen-receptor-mediated lymphocyte activation through the NF-κB pathway. Activation of NF-κB is essential for the immune system to function normally. Deregulated NF-κB in lymphocytes can lead to immunodeficiency, but also to autoimmunity or lymphomas. Phosphorylation of the CARMA1 and BCL10 proteins and ubiquitylation of BCL10 affect the formation and stability of the CARMA1-BCL10-MALT1 (CBM) complex, and also control negative feedback regulation of the NF-κB signaling pathway. Also, recent studies point out an unexpected role for BCL10 in the regulation of the actin cytoskeleton.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Structure
233 amino acids with the predicted molecular weight of approximately 26kD with the characteristic of caspase recruitment domain.
Distribution

BCL10 is expressed in all normal and malignant tissues. In the lymphoid tissue, it is highly expressed in the germinal center but low in the mantle zone, and intermediate in the marginal zone.
