No. 570104) (panel B) for 6 hours, were fixed with fixation buffer (Cat. 420801) for 10 minutes, permeabilized with 0.5% Triton X-100, and blocked with 5% FBS for 1 hour at room temperature.     Direct ELISA showing Purified anti-CD54 (ICAM-1) (clone W21128C) bound to plate-immobilized recombinant human ICAM-1-Fc Chimera protein (Cat. Nos. 552904 and 552906). The wells were incubated with serially diluted primary antibody at room temperature for 2 hours in 1% BSA in PBS, followed by incubation with HRP Goat anti-rat IgG (Cat. No. 405405). TMB High Sensitivity Substrate Solution (Cat. No. 421501) was used for detection. Absorbance was measured at 450 nm.

Description

Intercellular adhesion molecule 1, also known as ICAM-1 or CD54, is a member of the immunoglobulin superfamily. Structurally, ICAM-1 is characterized by heavy glycosylation with an extracellular domain composed of multiple loops created by disulfide bridges within the protein. Beta sheets are the dominant secondary structure of this protein, suggesting the presence of dimerization domains in ICAM-1. This protein is constitutively expressed at low levels on the cell surface of leukocytes, endothelial cells, macrophages, and lymphocytes. Upon cytokine activation, by cytokines such as IL-1 and TNFα, the concentration of ICAM-1 on the cell surface greatly increases. The ligand of ICAM-1 is LFA-1 (leukocyte function associated antigen 1), which is found on all T cells. LFA-1/ICAM-1 interaction has been shown to be important for T cell-T cell interactions, which leads to further T cell differentiation. ICAM-1 was found to play a role in infectious diseases, as it was identified as the binding target for entry of the major group of human rhinoviruses into various cell types and is also known for its affinity for Plasmodium falciparum-infected erythrocytes. These findings indicate that ICAM-1 may participate in the pathogenesis of several infectious agents.

Recommended Usage

Each lot of this antibody is quality control tested by immunocytochemistry. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/mL is recommended. For Direct ELISA, the optimal concentration of this reagent for the specific application and the experiment design should be established by the end-user. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For immunocytochemistry (ICC), 4% PFA fixation followed by permeabilization with 0.5% Triton X-100 or ice-cold methanol or fixation with ice-cold methanol is recommended.

MUC1 and promotes cell aggregation in epithelial cells, ARHGEF26/SGEF, (on T cell side) with CD81, CD247 and CD9 at immunological synapses between antigen-presenting cells and T cells, major receptor group rhinovirus A-B capsid proteins, and Coxsackievirus A21 capsid proteins.
