IHC staining using purified anti-COMT (clone A17180E) on formalin-fixed paraffin-embedded human placenta tissue. The tissue was incubated with 5 μg/mL of anti-COMT antibody overnight at 4°C, followed by incubation with 2.5 μg/mL of Alexa Fluor® 647 goat anti-mouse IgG (red) (Cat. No. 405322) for one hour at room temperature. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801), and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40x objective. Scalebar = 50 μM

    HeLa cells were fixed with 100% ice-cold methanol for 10 minutes and blocked with 5% FBS for 60 minutes. Cells were then intracellularly stained with either 5.0 µg/mL (1:100 dilution) purified mouse IgG1, κ isotype control (Cat. No. 401402) (panel A) or 1.25 µg/mL purified anti-COMT (clone A17180E) (panel B) overnight at 4 °C, followed by incubation with Alexa Fluor® 594 goat anti-mouse IgG (Cat. No. 405326) at 1:200 dilution. Nuclei were counterstained with DAPI and the image was captured with a 60X objective. Scale bar = 50 μm

Description

Catechol-O-methyltransferase (COMT) catalyzes the addition of methyl groups to catecholamine hormones and neurotransmitters; in particular: dopamine, norepinephrine, and epinephrine. Norepinephrine and epinephrine are key mediators of the sympathetic nervous system, while dopamine is an important neurotransmitter in the regulation of mood and behavior. Since O-methylation inactivates target catecholamines and leads to its degradation, COMT activity has significant implications for human health. The Val158Met polymorphism of the COMT gene is associated with mental disorders such as schizophrenia, obsessive compulsive disorder, bipolar disorder, and substance dependence.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 5 - 10 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 2.5 μg/mL is recommended. For intracellular flow cytometric staining, the suggested concentration range of this reagent is 0.01 - 0.03 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Product is not recommended for Western Blot. 

For ICC, recommend fixation/permeabilization with 100% ice-cold methanol. Do not recommend fixation with 4% PFA.
