Total lysates (15 µg protein) from HEK293T and HEK293T/FOXO3 CRISPR/Cas9 knockout (KO) cells were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to nitrocellulose, and probed with 1:500 diluted (1.0 µg/mL) purified anti-FOXO3 antibody, clone W15111A. Proteins were visualized by chemiluminescence detection using HRP-conjugated goat anti-mouse-IgG secondary antibody (Cat. No. 405307) at a 1:3000 dilution. Description

The Fork-head box O (FOXO) family of transcription factors is comprised of four family members; FOXO1, FOXO3, FOXO4, and FOXO6. FOXO3 was originally identified as a tumor suppressor and has been shown to regulate several cellular processes, such as cell cycle arrest, apoptosis, autophagy, longevity, and stress response. FOXO3 activity is determined by its post-translational modifications and subcellular localization. FOXO3 is inactivated by cytoplasmic retension through AKT/PI3K signaling pathways. FOXO3 inactivation has been associated with cell proliferation and tumorigenesis. In contrast, over-expression of FOXO3 leads to inhibition of tumor growth. FOXO3 also plays an important role in the development of regulatory T cells. FOXO3-deficient mice showed organ inflammation phenotype and increased production of proinflammatory cytokines during viral infection.
