HEK293 transiently transfected with CLEC4D-FLAG and FcRg chain cells were fixed with 2% paraformaldehyde (PFA) for twenty minutes and blocked/permeabilized with 10% FCS/0.05% saponin in PBS for one hour. Then the cells were stained with 5 µg/ml purified anti-human CD368 (clone 9B9) in 2% FCS/0.05% saponin in PBS for one hour at room temperature, followed by 2 µg/ml Alexa Fluor® 488 rabbit anti-mouse IgG (green) in 2% FCS/0.05% saponin for ten minutes at room temperature. Nuclei were counterstained with DRAQ5™ (blue). The image was captured with a 63X objective. Image provided by Jenny Ostrop of the Norwegian University of Science and Technology.

Description

CD368 (CLEC4D), also known as CLEC6, is a 30 kD single pass type II transmembrane protein and member of the C-type lectin-like domain (CTLD) superfamily. CLEC4D is an endocytic receptor, probably involved in antigen presentation, cell adhesion, and cell signaling. CLEC4D is expressed by macrophages, monocytes, neutrophils, and a subset of dendritic cells and is upregulated by IL-6, IL-10, TNF-α, and IFN-γ.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 1.0 µg per million cells in 100 µl volume. It is recommended that the reagent be titrated for optimal performance for each application.
