Whole cell extracts (15 µg total protein) from the indicated cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL purified anti-Merlin antibody (clone W19231A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin antibody (Cat. No. 643807) was used as a loading control at a 1:10000 dilution (lower). Lane M: Molecular weight marker.

    Whole cell extracts (250 µg total protein) prepared from HeLa cells were immunoprecipitated overnight with 2.5 µg of purified rat IgG2b, κ isotype ctrl antibody (Cat. No. 400602) or purified anti-Merlin antibody (clone W19231A). The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with a control antibody raised against a separate epitope of Merlin. Description

Moesin-ezrin-radixin-like protein (Merlin) is a tumor suppressor protein encoded by the Neurofibromin 2 (NF2) gene. It functions as a scaffold linking the plasma membrane and the cytoskeleton, and plays a crucial role in regulating cell polarization and morphology. Merlin functions as a major upstream activator of the Hippo signaling pathway, where it promotes Lats1/2 degradation, leading to increased nuclear localization of the YAP/TAZ transcription factors. Mutations and deletions within the NF2 gene cause neurofibromatosis type 2, a tumorigenic disease of the brain.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.5 - 2.0 µg/mL. For immunoprecipitation, the suggested use of this reagent is 2.5 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone was tested for ICC using 4% PFA-fixed HeLa cells permeabilized with either methanol or Triton X-100, and methanol-fixed and permeabilized HeLa cells. None of the methods were compatible with Merlin staining.

This clone is not predicted to cross-react with other Merlin homologs due to poor sequence conservation between the immunizing peptide and the corresponding region in each homolog.
