Cell lysate from Ramos (human, lane 1), Raji (human, lane 2), HeLa (human, lane 3), EL4 (mouse, lane 4) and A20 cell lines (mouse, lane 5) (15 µg protein for each) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to nitrocellulose, and probed with 1 µg/mL of Purified anti-NTAL Antibody (clone NAP-07) (upper). Proteins were visualized using chemiluminescence detection after incubation with a 1:3000 dilution of HRP conjugated anti-mouse-IgG secondary antibody for the anti-NTAL antibody (upper panel) or a 1:5000 dilution of Direct-Blot™ HRP anti-β-Actin (clone 2F1-1, lower panel) Antibody. Lane M: Molecular weight ladder.

Description

Non-T cell activation Linker (NTAL) is a transmembrane adaptor protein with an apparent molecular weight of 30 kD expressed on B cells, natural killer (NK) cells, and myeloid cells. The NTAL protein is found in rafts, which are membrane microdomains that participate in B cell activation. NTAL interacts with a variety of proteins including Grb2, PLC gamma 2, and PLC gamma 1. This gene is one of the contiguous genes at 7q11.23 commonly deleted in Williams syndrome.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. Western blotting, suggested working dilution(s): Use 10 µl per 5 ml antibody dilution buffer for each mini-gel. For IHC, use a 15 µl/ml dilution of antibody for staining. Antigen retrieval for IHC of formalin-fixed paraffin-embedded tissue using 0.01 M sodium citrate buffer is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

NOTE: For flow cytometric staining with this clone, True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401) offers improved staining and is highly recommended.

Antigen Details

Structure
30 kD double palmitoylated protein gene that consists of 14 exons, and its alternative splicing generates three transcript variants.
Distribution

B, natural killer cells, monocytes, mast cells, and activated T cells.

Function
Involved in FCER1 mediated signaling in mast cells and as a negative regulator of early B cell signaling. May also be involved in CFCGR1 mediated signaling in myeloid cells. Couples activation of these receptors with distal intracellular events through the recruitment of Grb2.
Ligand/Receptor
Membrane microdomains (rafts) of B cells, natural killer (NK) cells, and myeloid cells.
Cell Type
B cells, NK cells, Monocytes, Mast cells, T cells
Biology Area
Cell Biology, Immunology, Signal Transduction
