    Synthetic human β-Amyloid peptides 1-38 (blue circles), 1-40 (Cat. No. 932601) (red squares), or 1-42 (Cat. No. 932501) (green triangles) were coated onto Costar™ 96-well, high binding assay plate at 2 µg/mL and then incubated with a dilution series of purified anti-β-Amyloid, 1-38 recombinant antibody (clone QA19A28). Bound antibodies were detected with HRP donkey anti-rabbit IgG antibody (Cat. No. 406401) followed by TMB substrate solution. Absorbance was measured at 450 nm.

    Synthetic β-Amyloid peptides 1-38, 1-40, and 1-42 (1 µg per lane) were run on a 16.5% Tris-Tricine gel, transferred to Nitrocellulose, and probed with 0.5 µg/mL of anti-β-amyloid, 1-38 recombinant antibody overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rabbit IgG antibody (Cat. No. 406401) at a 1:3000 dilution. Lane M: Molecular weight marker.

Description

Amyloid beta (Aβ or Amyloid beta) denotes peptides of 36-43 amino acids in length that are crucially involved in Alzheimer's disease as the main component of the amyloid plaques found in the brains of Alzheimer patients. The peptides result from the amyloid precursor protein (APP), which is cut by certain enzymes to yield Aβ. Aβ molecules can aggregate to form oligomers (known as "seeds") which are believed to be able to induce other Aβ molecules to also take the misfolded oligomeric form, leading to a chain reaction akin to a prion infection. The seeds or the resulting amyloid plaques are toxic to nerve cells. The other protein implicated in Alzheimer's disease, tau protein, also forms such prion-like misfolded oligomers, and there is some evidence that misfolded Aβ can induce tau to misfold.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 2.5 - 10 µg/mL is suggested.  For immunohistochemistry, a concentration range of 5.0 - 10 µg/mL is suggested. Recommended usage concentration for direct ELISA application is 29 ng/mL. For western blotting, the suggested use of this reagent is 0.5 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody is not recommended for WB application. It specifically recognizes β-amyloid peptide 1-38 but has poor sensitivity against the target.
