IHC staining of purified anti-Parvalbumin (clone A17130G) on formalin-fixed paraffin-embedded mouse cerebellum tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., 1X (Cat. No. 928502), the tissue was incubated without (panel A) and with (panel B) 5 µg/mL of purified anti-Parvalbumin (clone A17130G) followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 10X objective. Scale bar: 50 µm

Description

Parvalbumin is a calcium-binding protein with diverse functions in various tissues. Originally identified in skeletal muscle fibers, parvalbumin holds particular significance in the central nervous system. In neurons, parvalbumin acts as a calcium buffer, helping to maintain precise control over calcium ion levels within the cell. This function is critical for regulating synaptic transmission, especially at inhibitory synapses where it influences the release of the neurotransmitter GABA to synchronize the activity of neural networks. Additionally, parvalbumin contributes to the plasticity of synaptic connections, affecting learning and memory. IHC-P - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For IHC-P in mouse tissues, we recommend antigen retrieval with Citrate Buffer, 10X (Cat. No. 420901). 

Clone A17130G is not suitable for Western Blot.

Clone A17130G is not suitable for ICC. 

Clone A17130G is not suitable for IHC-P in human tissues. Although signal is detected, signal strength is low.
