IHC staining of purified anti-SMYD2 (clone 1G10-E11-H9) on formalin-fixed paraffin-embedded human kidney tissue. Following antigen retrieval using Tris EDTA, pH 9.0, the tissue was incubated with 10.0 µg/mL of the primary antibody overnight at 4°C, followed by incubation with Alexa Fluor® 647 goat anti-mouse IgG (Cat. No. 405322) for one hour at room temperature. Nuclei were counterstained with DAPI and the slides were mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40X objective. Scale bar: 50 µm

Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.25 µg/mL of purified anti-SMYD2 (clone 1G10-E11-H9) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin (Cat. No. 664804) was used as a loading control at a 1:25000 dilution. Lane M: Molecular weight marker

Description

SET and MYND domain-containing protein 2, also known as SMYD2, is one of five members of the SMYD family of lysine methyltransferases composed of an evolutionarily conserved SET domain and a zinc finger-containing MYND domain. SMYD2 localizes to the nucleus and cytoplasm and methylates histones such as H2B, H3K4, H3K36, and H4 as well as non-histone substrates, notably p53 on lysine 370. As a protein lysine methyltransferase, SMYD2 is a regulator of transcription and epigenetic signaling and functions in the regulation of cell cycle and apoptosis. It has also been shown to promote NF-κB signaling pathways via methylation of TRAF2 and STAT3. Aberrant SMYD2 expression is associated with a variety of cancers and other diseases including kidney disease, cardiovascular disease, and rheumatoid arthritis. It promotes tumorigenesis and metastasis through the repression of tumor suppressor proteins RB1, p53, and PTEN.
