Lane 1, 5: Molecular weight marker; Lane 2: 2 µg of human liver lysate; Lane 3: 20 µg of HepG2 lysate; Lane 4: 20 µg of MCF7 cell lysate. The blot was incubated with 0.2 µg/mL primary antibody overnight at 4°C, followed by incubation with HRP goat anti-mouse IgG antibody (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system. Cell lysates were loaded in order of descending EGFR expression; predicted expression data was obtained from Human Protein Atlas.

    ICC staining of purified anti-Fibrinogen antibody (clone G8 1-2) on HepG2 cell. The cells were fixed with 4% PFA, permeabilized with a buffer containing 0.1% Triton X-100 and 0.25% BSA, and blocked with 2% normal goat serum and 0.02% BSA. The cells were then incubated with 5 µg/mL primary antibody overnight at 4°C, followed by incubation with 2.5 µg/mL Alexa Fluor® 594 goat anti-mouse IgG for one hour at room temperature. The cells were co-stained with Flash Phalloidin™ Green 488 (Cat. No. 424201). The slide was mounted with fluoromount G with DAPI. The image was captured with a 60X objective. Scale bar: 20 µm.

Description

Fibrinogen is a glycoprotein that is synthesized by hepatocytes in the liver. Fibrinogen alpha chain (FGA) is the alpha component of the fibrinogen, which is released to plasma after fibrinogen is cleaved by thrombin. Upon vascular injury, FGA forms the fibrin complex with fibrinogen beta (FGB) and fibrinogen gamma (FGG). The complex is one of the primary components of blood clots.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.2 - 1.0 µg/mL.  For immunocytochemistry, the suggested use of this reagent is 1.0 - 5.0 μg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
