Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 μg/mL of purified anti-human ALK (clone W16190A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Lane M: Molecular weight marker

Description

ALK is expressed widely in the central nervous system, where it plays important roles in neurogenesis and the development of the nervous system. ALK is a receptor tyrosine kinase that phosphorylates tyrosines on target proteins upon ligand binding (known ligands include pleiotrophin, midkine, FAM150A, and FAM150B). ALK was originally discovered in anaplastic large-cell lymphoma cells where a chromosomal translocation leads to constitutive activation of the ALK kinase domain, making ALK a target of inhibition in cancer therapies.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.5 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

W16189A is not suitable for ICC, FC, and IHC-P.

W16189A is weakly cross-reactive with mouse ALK in WB.
