Total cell lysate from human HeLa cells (lane 1), U87-MG cells (lane 2), and RPMI8226 cells (lane 3) were resolved by electrophoresis, transferred to nitrocellulose, and probed with purified monoclonal anti-P2RX7 (clone W15180A) antibody. Proteins were visualized using an HRP Goat anti-mouse IgG Antibody and chemiluminescence detection. Purified anti-GAPDH (clone Poly6314) antibody was used as a loading control.

Description

P2X purinoceptor 7 (P2RX7) is a member of the purinergic receptor P2X family. P2 purinergic receptors are cell surface receptors that are involved in proliferation of neural stem cells and have been implicated in learning, memory, and sleep. P2X receptors are ligand-gated ion channels that mediate calcium and potassium fluxes in response to ATP binding. Functional P2X receptors are homo- or heterotrimeric complex composed of the P2X family members. P2RX7 functions as a ligand-gated cation channel and is responsible for synaptic transmission and ATP-dependent lysis of macrophages through the formation of membrane pores. P2RX7 is highly expressed in hematopoietic cells and is involved in cell death, host defense against pathogens, and the regulation of inflammatory responses. P2RX7 is also found in the nervous system, microglia, uterine endometrium, and the retina. Mutations in the P2RX7 gene have been associated to major depressive disorder (MDD) and low bone mineral density.
