Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 1 µg/mL (1:500 dilution) of purified anti-PTK2B (clone W20075C) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL substrate premium kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

  • B_W20075C_PURE_PTK2B_Antibody_ICC_120122
    ICC staining of purified anti-PTK2B (clone W20075C) on Daudi cells. The cells were fixed and permeabilized with iced-cold 100% methanol, and blocked with 5% FBS for 1 hour at room temperature. The cells were then stained with 5.0 µg/mL of the primary antibody, followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 Goat anti-rat IgG (red) (Cat. No. 405422) for 1 hour at room temperature. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801). Scale bar: 20 μm

Description

Protein-tyrosine kinase 2-beta (PTK2B or PYK2), a calcium-dependent non-receptor tyrosine kinase, belongs to focal adhesion kinase family and shares approximately 47% amino acid sequence with FAK (or PTK2). As a member of focal adhesion family, PTK2B’s phosphorylation enables interactions with other scaffold and effector proteins that regulate cell migration, adhesion, spreading, actin cytoskeleton reorganization and focal adhesion contact. PTK2B responds to several cellular stimuli to regulate critical phycological functions such as G-protein coupled receptor stimulation in vascular muscle cells, T cell and B cell antigen receptor engagement, and calcium-activated MAP kinase signaling pathway in neuronal cells. A genome-wide research suggests PTK2B correlates with Alzheimer’s disease and is able to reduce the Tau toxicity in animal models.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This product does not cross react with mouse in western blotting. This product is verified to work in immunocytochemistry with methanol only fixed cells but not paraformaldehyde-fixed cells. This product is not suitable for IHC-P (fluorescence).
