Recombinant Human TNFSF18 (GITRL) chimera (Cat. No. 559204) (black circles) binds to immobilized human GITR (TNFRSF18) Fc chimera (Cat. No. 764302). Ultra-LEAF™ purified anti-human TNFSF18 (GITRL) (clone W21092F) (purple squares) inhibits the binding in a dose-dependent manner, whereas the Ultra-LEAF™ purified rat IgG2a, κ isotype control (Cat. No. 400544) (gray triangles) does not have the effect. This antibody blocks the binding of 0.15 µg/mL recombinant human TNFSF18 (GITRL) to 2 µg/mL immobilized recombinant human GITR (TNFRSF18) Fc chimera. ND50 range: 0.12 - 1 µg/mL.

Description

TNFSF18 is a type II transmembrane glycoprotein that belongs to the TNF superfamily. This protein is mainly expressed on antigen presenting cells (APC) including macrophages, B cells, dendritic cells (DCs), and endothelial cells. Its expression increases upon the stimulation of proinflammatory stimuli. The receptor of TNFSF18 is the glucocorticoid-induced TNFR-related protein (GITR), which is a member of the TNFR superfamily. Engagement of TNFSF18 and GITR leads to T cell activation, proliferation, and cytokine production. It also inhibits the suppressive function of T regulatory cells and induces macrophage activation. GITR-TNFSF18 interaction plays an essential role in the regulation of the immune response to self-antigens and pathogens. In addition, this receptor-ligand interaction has been shown to modulate anti-tumor immunity.

Block - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by blocking the binding of 0.15 µg/mL human TNFSF18 (GITRL) to 2 µg/mL immobilized recombinant human GITR (TNFRSF18) Fc chimera. ND50 range: 0.12 – 1 mg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Related FAQs

Do you guarantee that your antibodies are totally pathogen free?

BioLegend does not test for pathogens in-house aside from the GoInVivo™ product line. However, upon request, this can be tested on a custom basis with an outside, independent laboratory.

Does BioLegend test each Ultra-LEAF™ antibody by functional assay?

No, BioLegend does not test Ultra-LEAF™ antibodies by functional assays unless otherwise indicated. Due to the possible complexities and variations of uses of biofunctional antibodies in different assays and because of the large product portfolio, BioLegend does not currently perform functional assays as a routine QC for the antibodies. However, we do provide references in which the antibodies were used for functional assays and we do perform QC to verify the specificity and quality of the antibody based on our strict specification criteria.

Does BioLegend test each Ultra-LEAF™ antibody for potential pathogens?

No, BioLegend does not test for pathogens in-house unless otherwise indicated.  However, we can recommend an outside vendor to perform this testing as needed.

Have you tested this Ultra-LEAF™ antibody for in vivo or in vitro applications?

We don't test our antibodies for in vivo or in vitro applications unless otherwise indicated. Depending on the product, the TDS may describe literature supporting usage of a particular product for bioassay. It may be best to further consult the literature to find clone specific information.
