Biotinylated Human LAIR2-Fc Chimera was aliquoted in PBS, pH 7.2 at 0.2 mg/mL and one aliquot was kept at 4°C (Control), and another was frozen and thawed four times (4x Freeze/Thaw). After this procedure, the samples were tested for their ability to bind to collagen type I in a dose-dependent manner. The ED50 for this effect is 0.175 - 1.4 ng/mL.

Description

LAIR2 (leukocyte associated immunoglobulin like receptor 2) is a member of the immunoglobulin superfamily and a natural soluble homolog of LAIR1 that is present in humans and non-human primates, but not other animals. LAIR2 shares 84% identity with the LAIR1 extracellular region, binds to collagens with higher affinity than LAIR1 and functions as a natural antagonist to block cell membrane LAIR1 inhibitory signaling. This gene maps to a region of 19q13.4 in human chromosome, termed the leukocyte receptor cluster, which contains 29 genes in the immunoglobulin superfamily. LAIR2 may play roles in both inhibition of collagen-induced platelet aggregation and vessel formation during placental implantation. Amount of soluble LAIR1 and LAIR2 are significantly increased in urine samples of RA patients and significantly correlated with the markers of inflammation. In vitro stimulation of sorted peripheral blood cells revealed that CD4+ T cells are the main producers of LAIR2. LAIR1 and LAIR2 bind to the collagen-like regions of C1q and of MBL (mannose binding lectin) and can efficiently inhibit complement activation via classical and lectin pathways. LAIR2 could inhibit tumor metastasis, invasion, and collagen deposition via suppressing TGF-β signaling suggesting that LAIR2 might act as a tumor suppressor.

Biotinylated human LAIR2-Fc Chimera, amino acids Gln22-Pro152 (Accession # Q6ISS4), with a C-terminal Fc and Avi tags, was expressed in 293E cells. Human LAIR2-Fc-Avi tag was site-specifically biotinylated by enzyme BirA.
Molecular Mass
The 391 amino acid recombinant protein has a predicted molecular mass of approximately 43.0 kD. The DTT-reduced and non-reduced protein migrates at approximately 45-55 kD and 75-85 kD respectively in SDS-PAGE. The predicted C-terminal amino acid is Gln.
Purity
> 95%, as determined by Coomassie stained SDS-PAGE.

Endotoxin Level
Less than 0.1 EU per µg of cytokine as determined by the LAL method.
Concentration
25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at - 20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
