Total cell lysates (15 µg protein) from the indicated cell lines were resolved by electrophoresis (4-12% Bis-Tris gel), transferred to a nitrocellulose membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-TCF8 (ZEB1) Antibody, clone P82A8A6. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse-IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-Actin Antibody (Cat. No. 664804) was used as a loading control at a 1:5000 dilution. Lane M: Molecular weight ladder.

Description

TCF-8, also named ZEB1, belongs to the ZEB family of transcription factors characterized by the presence of 2 zinc finger clusters and are responsible for the binding to specific DNA sequences named E-boxes. By recruiting co-suppressors or co-activators through CID, SID or CBD, TCF-8 can either downregulate or upregulate the expression of its target genes.

TCF-8 promotes tumor invasion and metastasis by inducing epithelial-mesenchymal transition (EMT) in carcinoma cells. It is associated with aggressive behaviour, metastasis, treatment resistance and poor prognosis in different tumour types, including breast, pancreatic, prostate and lung cancer.
