IHC staining of formalin-fixed paraffin-embedded human cerebellum stained with purified anti-TRIP8B (clone N212-17). Following antigen retrieval using 0.1 M Tris-buffered saline with Tween 20 (Cat. No. 925501), the tissue was incubated with 5 µg/mL of the primary antibody overnight at 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 647 goat anti-mouse IgG (red) (Cat. No. 405416) for one hour at room temperature. Nuclei were counterstained with DAPI (blue), and the slide was mounted with ProLong&tradel Gold Antifade Mountant.     Whole cell extracts (15 µg total protein) from human, rat, and mouse brain membrane lysates were heated at 50°C for 10 minutes, resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 μg/mL of purified TRIP8b antibody (clone N212/17) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular weight marker.

Description

TRIP8b is a protein that binds to and regulates hyperpolarization-activated cyclic nucleotide-gated (HCN) channels, an important class of voltage-gated channels. Binding of TRIP8b to HCN channels causes them to open at more hyperpolarized potentials, as TRIP8b antagonizes the binding of cyclic nucleotides. Additionally, TRIP8b affects HCN channel subcellular localization. Via these functions, TRIP8b plays a role in learning and memory, major depressive disorder, and epilepsy.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 5.0 - 10.0 µg/mL is suggested. For western blotting, the suggested use of this reagent is 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Does not react with human in Western blot. Lysates were heated at 50°C for ten minutes prior to electrophoresis.
