Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 1 µg/mL (1:500 dilution) of purified anti-β-Tubulin recombinant (clone QA20A98) overnight at 4°C.     IHC staining using purified anti-β-Tubulin recombinant (clone QA20A98) on formalin-fixed paraffin-embedded human brain tissue. Following antigen retrieval using 1X Tris-Buffered Saline (final concentration 0.05M) with Tween-20 (Cat. No. 925501), the tissue was incubated with 10 μg/mL of antibody overnight at 4°C, followed by incubation with 2.5 μg/mL of Alexa Fluor® 647 goat anti-mouse IgG (Cat. No. 405322) for one hour at room temperature. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801), and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40x objective. Scale bar = 50 μM

The cells were fixed and permeabilized with 4% paraformaldehyde, permeabilized with 0.5% Triton X, and blocked with 5% FBS for 1 hour at room temperature. The cells were then stained with 1.0 µg/mL of the primary antibody, followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 goat anti-mouse IgG (Cat. No. 405326) for 1 hour at room temperature. Description

Eukaryotic microtubules are tubular polymers of tubulin that are a major component of the cytoskeleton. They play an essential role in diverse cellular functions, including motility, mitosis, and trafficking, and are also critical for normal development. β- and α-tubulins comprise the two core protein families that heterodimerize and assemble into each microtubule filament. There are multiple β-tubulin isotypes, with each showing unique tumoral and tissue-specific distribution.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For ICC, staining was compatible with fixation with 100% ice-cold methanol, or fixation with 4% paraformaldehyde followed by permeabilization with 100% ice-cold methanol or 0.5% Triton-X. 

This antibody is not designed to be specific against any particular isoform of β-tubulin.
