    Total cell lysate (300 µg protein) from NIH3T3 were incubated with 2.5 µg mouse IgG1 isotype control (lane 1) or anti-IκB-α antibody (lane 2) at 4°C for 3 hours, followed by incubating the Protein-G beads overnight. Immunoprecipitates and total protein (10% of input, lane 3) were eluted with SDS loading buffer and resolved by 4-12% Bis-Tris gel electrophoresis, transferred to nitrocellulose, and probed with 1:500 (1 µg/mL) diluted purified anti- IκB-α antibody (clone W16166B, upper). Proteins were visualized using 1:3000 diluted HRP goat anti-rat IgG secondary antibody (Cat. No. 405405) and chemiluminescence detection. Lane M is the MW ladder. Description

IκB-α is an inhibitory protein that binds to the NF-κB complex and retains this complex in the cytoplasm where it cannot activate transcription. Upon cellular stimulation, the IκB proteins are phosphorylated and degraded via the ubiquitin pathway allowing for nuclear translocation of NF-κB. The NF-κB complex can then bind DNA to activate gene expression of multiple proteins involved in inflammation, cellular proliferation, differentiation, and apoptosis.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.125-1.0 µg per ml (dilution of 1:500-1:4000). For immunoprecipitation, the suggested use of this reagent is 2.5 - 3.00 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Weakly cross reacts with human I?Ba.

This clone can not be used for ICC application.
