IHC staining of purified anti-HSF1 antibody (clone 4B4) on formalin-fixed paraffin-embedded human bladder tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., the tissue was incubated with 10 µg/mL of the primary antibody overnight at 4°C. Following a biotinylated goat anti-rat linking reagent, BioLegend’s Ultra-Streptavidin (USA) HRP labeling reagent and DAB was used for detection followed by hematoxylin counterstaining, according to the protocol provided. Description

HSF1 is a DNA-binding protein that exists as an inactive monomer in a complex with Hsp40/Hsp70 and Hsp90.  Upon stress, HSF1 is released from the chaperone complex and trimerizes.  HSF1 is then transported into the nucleus where it is hyperphosphorylated and binds to DNA containing heat shock elements (HSE) and activates transcription of proteins such as HSP70.  HSP70 is known to inhibit the tau and alpha synuclein aggregation seen in Alzheimer’s Disease and Parkinson’s Disease.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
