Total cell lysates (15 µg total protein) from Saos-2, U937 (reduced expression control), THP-1 (negative control), and THP-1(reduced expression control) cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 0.005 µg/mL (1:100,000 dilution) of Purified anti-RUNX2 Antibody, clone W15212A, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG Antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50,000 dilution (lower). Lane M: Molecular Weight marker. Predicted expression data was obtained from Human Protein Atlas.

Description

Runt Related Transcription Factor 2 (RUNX2), also known as CBFA1, belongs to the mammalian Runt-related transcription factor (RUNX) family. The RUNX family is comprised of three members: RUNX1, RUNX2, and RUNX3. The RUNX proteins are DNA-binding proteins that regulate the expression of genes involved in cellular differentiation and cell cycle progression. Among these members, RUNX2 contains one Runt domain and specifically expresses in osteoblasts. It plays critical roles in skeletal mineralization, osteoblast differentiation of mesenchymal stem cells, chondrocyte hypertrophy, endothelial cell migration, and vascular invasion of developing bones. Defects in this gene have been associated with the bone development disorder cleidocranial dysplasia (CCD) and defective endochondral and intramembranous bone formation. In some cases, defects in RUNX2 are exclusively associated with dental anomalies. RUNX2 binds DNA as a monomer or as a subunit of a heterodimeric complex. In osteoblasts, RUNX2 supports transcriptional activation by synergizing with SPEN/MINT. RUNX2 also functions to inhibit KAT6B (MYST4)-dependent transcriptional activation.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.002 - 0.01 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.
